Fab and F(ab’)2 Preparation Kits - CD Bioparticles Antibody fragments have been widely used in a variety of disciplines ranging from bioanalytical immunoassays to pharmacological research. They are with small size and lack an Fc-mediated binding region allowing for better penetration into tissue and various applications. There are two primary ways to produce such fragments: One is digestion with an enzyme such as papain for Fab fragments, pepsin for F(ab')2 fragments, or ficin for mouse IgG1 Fab and F(ab')2 which are difficult to cleave with papain and pepsin. The other is expression of a recombinant antibody fragment. Especially, enzymatic digestion can rapidly produce fragments from commercially available antibodies. Although enzymatic antibody fragmentation with pepsin has been studied since the 1960's and 70's, most laboratories still must optimize the fragmentation of each particular antibody by trial and error.
